Fig.1 Detection of IL15RA expression in mammary gland and spleen by RT-PCR. Mouse Il15ra mRNA (120 bp) was detectable only in wild-type C57BL/6 mice. Human IL15RA mRNA (124 bp) was detectable only in homozygous hIL15RA knockin mice but not in wild-type mice. Mammary gland and spleen RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=2) and homozygous hIL15RA knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Leukocytes were collected from wild-type C57BL/6 mice (male, 6 weeks old) and homozygous HO hIL15RA humanized mice (male, 6 weeks old),stimulated with LPS in vitro(1ug/ml), and analyzed by flow cytometry with anti-mIL15RA , anti-hIL15RA.
